Influence of Excess of Antigen on Immunological Tolerance in Radiation Chimæras

When the lymphatic system of an irradiated allogenic animal is repopulated with cells from normal bone marrow, tolerance may only develop if sufficient antigens are present. Transplantation of even a small number of normal lymph node cells always results in an immunological graft versus host reaction and a killing effect'' of the host. Transfer of bone marrow from a tolerant chimaera to an irradiated animal in which the specific antigens are absent results in a loss of this tolerance, and when excess of another antigen is present, tolerance to this antigen may develop. A factor that determines the success of the transfer of tolerance is the dose of lymph node cells. Their number should be far in excess of the bone marrow cell dose. A competition to repopulate the lymphatic tissue seems to exist between bone marrow and lymphoid cells. These results indicate that mouse bone marrow is a source of cells with undetermined immunological function. These cells can develop into immunologically active lymphoid cells or into tolerant lymphoid cells when an antigen excess is present.
