HISTOLOGIC AND HISTOCHEMICAL CHANGES IN RABBIT KIDNEY AFTER ROENTGEN IRRADIATION

The kidney of rabbits was irradiated with 5000 r and examined 18 hr to 46 days later. Relatively slight changes were noted during the early postirradiation period when routine histologic techniques were used. At this period, hyperemia was the principal finding. After 5 to 7 days, elastic fibers of blood vessel walls were fragmented and interstitial reticulin fibers were swollen and stained poorly with silver. Histochemical methods showed a depletion of sulfhydryl and disulfide groups in parenchymal tissue. Esterase activity was low by the Sth day, then returned to normal. Lipase activity was increased from 18 hr to the 9th day, after which it declined. Pyruvic dehydrogenase showed a decrease 18 hr to 9 days after irradiation and a return to normal by the 18th day; changes in cytochrome oxidase were similar. Both alkaline and acid phosphatases showed an immediate rise in activity, which reached a max. on the 7th day. Then acid phosphatase fell sharply on the 9th day and alkaline phosphatase on the 12th day. Thereafter the activities of both enzymes rose gradually to normal levels. The histochemical results with these 2 enzymes were confirmed by chemical assays on kidney. Functional disturbances in kidney were reflected by increased urinary excretion of non-protein nitrogen after irradiation. This was seen morphologically at late postirradiation stages by fibrosis and loss of functioning tubules. It is suggested that the rise in lipase activity after irradiation resulted from increased demands for lipid transport in degenerating tissue. The impairment of activity of the respiratory enzymes (pyruvic dehydrogenase and cytochrome oxidase) indicates mitochondrial injury, which was evident at an early stage. (H.H.D.)
