The tumor pathology of thorotrast in man (in French)
An original survey of 22 new cases as well as an extensive review of previously published cases confirmed the carcinogenic action of Thorotrast. In 5 of these cases a primary carcinoma of the liver or biliary tract was present, 3 epitheliomas and 2 endothelial sarcomas. This is clearly a much higher level of hepatic carcinoma than in the general male population (2%) or in males with hepatic cirrhosis (9.5% incidence). Thorotrast is taken up mainly by reticuloendothelial cells, so that experimental turaors resulting from injection of colloidal ThO2 are almost entirely sarcomas. In man, the radiation from ThO2 deposits may act directiy in the malignant transformation of glandular or epithelial cells; on the other hand it may induce neoplasia secondarily from a toxic effect or sclerotic action of a foreign body. However, the relative rarity of liver sarcomas in the absence of a history of exposure to Thorotrast suggests a direct effect of ThO2 radiation in the carcinogenesis of these tumors. Nevertheless, the possibility that it acts as a cocarginogen cannot be ruled out at this time. 100 references are cited.
