THE INFLUENCE OF ROENTGEN RADIATION ON OXIDATIVE PHOSPHORYLATION IN ISOLATED MITOCHONDRIA (in German)

Mitochondria were sedimented from homogenates of rat liver in either isotonic sucrose medium or in isotonic KClmannitol medium. Oxidative phosphorylation, or the P/O ratio, was greater in sucrose than in the other medium in the presence of various substrates. Irradiation of mitochondria suspended in sucrose with 200-kv x rays at doses between 750 and 40000 r had no effect on respiration, phosphorylation, or P/O ratio in the presence of various substrates of the citric acid cycle, glutamic acid, or deg hydroxybutyric acid. In the presence of alpha -ketoglutarate, respiration and phosphorylation increased approximates 10 to 20% after 40000-r irradiation but the P/O ratio was unaffected. Irradiation of mitochondria suspended in isotonic KClmannitol solution resulted in a fall in respiration, phosphorylation, and the P/O ratio in the presence of all of the substrates tested. The metabolic activities of unirradiated mitochondria remained high when stored in isotonic sucrose for 24 hr but fell sharply by that time in the 2nd medium. (H.H.D.)
