MUTAGENIC EFFECT OF C 14 AND H 3 LABELLED DNA PRECURSORS INJECTED INTO DROSOPHILA MELANOGASTER MALES

Male larvae were injected with adenine-C¹⁴, cytidine-H³, and thymidine-H³ to determine whlch would give the highest induced mutation rate with this level of commercially available radioactivity. With regard to sex- linked recessive lethals, all mutation frequencies were significantly higher in experiments in which radioactive chemicals had been injected instead of inactive precursors of DNA. The mutation frequency was highest after adenine-8-C¹⁴ was injected into larvae, but there was no statistical difference whether tritium was present as tritiated cytidine or tritiated thymidine. The overall hatchability for all broods was about 98%. There was no indication that radioactivity induced a higher dominant lethal mutation rate. To verify that the radiochemicals had reached the sperm by the technique used, it was established that sperm transmitted to females were radioactive. Variation in mutation frequency mutation frequency increased until 12 to 13 days after injection when the frequency reached a peak and then fell off again. The data suggest that sex- linked recessive lethals may be used as a measure of the mutagenic effect of radiochemicals of the type employed. (TCO)
