EFFECTS OF X RAYS ON MITOSIS OF MAMMALIAN CELLS CULTURED IN VITRO (in Italian)

Cells of strain RCP isolated from guinea pig kidney were irradiated at a rate of 150 r/min with 180-kv x rays to doses of 150, 300, and 450 r. Studies were made of variations of mitotic index and frequencies of nuclear damage after exposure. The earliest recognizable effect of irradiation was a transitory inhibition of mitotic division in cells that were about to divide at the time of irradiation. The mitotic depression and length of the mitotic delay were proportional to dosage. After mitotic inhibition a partial synchronization of the cell population occurred. At the same time a mitotic block was induced as a result of several metaphasic alterations. Chromosome bridges were quantitatively recorded in anaphasic, telophasic, and interphasic cells at different times after irradiation. Giant cell induction was one of the most obvious effects of irradiation and may be related to chromosome-induced damage. Several nuclear alterations, such as multipolar spindies and micronuclei, showed an incidence proportional to the radiation dose and were considered to be a consequence of giant cell formation. At 72 hr after 450 r, multipolar spindles and micronuclei were found in 20 and 12% of the cells, respectively, compared with incidences of 2% and <1% in unirradiated cells. (TCO)
