PATHOGENESIS OF POST-IRRADIATION GLOMERULOSCLEROSIS IN MICE

Renal glomerular lesions were studied histologically in LAF/sub 1/ mice given 690-rad whole-body x radiation. The lesion was manifested by the proliferation of large, irregular pleomorphic cells in the intercapillary space, and by the subsequent deposition of PAS-positive material in these areas and in the glomerular basement membranes. The lesion appeared after a latent period of about 20 weeks and progressed rapidly from that time. Kidneys of nonirradiated LAF/sub 1/ mice 18 to 24 months old showed the development of lesions morphologically similar to but less severe than those observed in the mice examined more than 20 weeks after irradiation. Proliferated intercapillary Cells in the glomerular lesion were much more bizarre and pleomorphic in the young irradiated mouse than in the senile nonirradiated mouse. It is suggested that the similar but less severe lesions seen in nonirradiated aged LAF/sub 1/ mice may imply that the irradiation lesion is a manifestation of a premature aging phenomenon in the kidney although acceieration of what may be a spontaneous form of glomerulitis must also be considered.
