Repopulation of the bone marrow and lymph nodes with donor-type cells was demonstrated in C57BL/Rij and (CBA/Rij x C57BL/Rij)F/sub 1/ mice that had been exposed to a lethal dose of total body x irradiation and treated with CBA bone marrow cells. No apparent symptom s of secondary disease were found in those chimeras that survived 100 days after irradiation and bone marrow transplantation. The same sensitivity to a graft-vs-host reaction was found in C57BL and F/sub 1/ mice and long-term CBA yields C57BL and CBA YY F/sub 1/ chiumeras, as tested by the killing effect of CBA lymph node cells. Therefore, no alteration in sensitivity to a graft-vs-host reaction occurred in long-term chimeras. CBA lymph node cells derived from long-term chimeras showed the same immunologcal activity against isoantigens of a third (RF) strain as CBA lymph node cells from normal CBA mice, but no killing effect of chimeric lymph node cells could be demonstrated in irradiated C57BL or (CBA X cal tolerance of the donortype lymphoid cells towards the host's isoantigens occurred in the long-term chimeras studied.
