MODIFICATION OF THE FOREIGN SPLEEN REACTION: AMELIORATION OF BLOOD AND TISSUE CHANGES

A comparison was made of histologic and hematologic changes in 2 groups of lethally irradiated mice implanted with parental-strain spleen cells. In 1 group the spleen cells were incubated with liver of the recipient strain in an attempt to prevent the celis from causing subsequent detrimental reactions in the recipients, and in the 2nd group untreated spleen cells were transferred. Inactivation of C57BL spleen cells by their incubation with recipient-type (C57BL x 101) F/sub 1/ liver inhibited the blood and tissue changes otherwise occurring when such cells were injected in sublethally irradiated (C57BL x 101) F/sub 1/ mice. In recipients of such inactivated cells, hemopoietic regeneration was enhanced and the granulomatous reaction in the lymphoid tissues was reduced. It is inferred that pre-exposure of the grafted cells to recipient cells, or their antigens, lessened their ability to react subsequently against the host. The observed inactivation process may involve alteration of the donor antibody- forming cells by an excess of recipient antigens present in the incubation tissue. (H.H.D.)
