NEW TECHNIQUE FOR LABELING TRIOLEIN BY I$sup 131$ AND Br$sup 8$$sup 2$

An improved method, which gives a highly labeled fat for alimentary absorption studies, is described. The method depends upon the reaction of the double bond of oleic acid with 1 molecule of BrI. BrI is prepared by the reaction of NaI/sup 131/ and Br/sup 82/ NH/sub 4/ in the presence of KHSO/sub 3/; the reacti on is practically complete after the reagents are allowed to stand in aqueous solution at room temperature for 30 min. Labeling is carried out by introducing an ethanol solution of triolein into the mixture, shaking, adding acetic anhydride, and decanting the upper acetic acid phase. The excess BrI is removed by treatment with an aqueous solution of a reducing agent consisting of equal parts (by weight) of KI and Na/sub 2/S/sub 2/O/sub 3/. Analysis of the labeled fat showed that 97% of the theoretical amount of BrI had reacted with the unsaturated fatty acid molecules. Besides the high degree of labeling obtained with the method, it avoids the use of organic solvents, which are difficult to remove from the fat and provide a less favorable starting product for human fat absorption studies. Purification of the labeled triolein and its analysis after separation by paper and column chromatography are described. The results showed that less than 1% of the initial radioactivity of Br/sup 82/-I/sup 131/-labeled triolein was lost after 3 weeks' storage. The small amount of free radioactivity in the preparations did not interfere with the results of biologic studies. (H.H.D.)
