THE CATABOLISM OF HEXOBARBITAL IN VIVO IN THE WHOLE-BODY IRRADIATED MOUSE

In mice x irradiated with 700 r, the catabolism of injected hexobarbital was significantly depressed when tested on the 5th and 7th postirradiation days. The respective elimination constants (calculated by multiplying the regression coefficient by 1/log e) were -0.0067 and -0.0059 min/sup -1/, in contrast to the value in unirradiated mice of --0.0148, which corresponds to a 59% decline in the concentration of hexobarbital 1 hr after injection. On the other hand, catabolism of hexobarbital was accelerated on the 8th and 14th days after 400-r wholebody irradiation; the corresponding elimination coefficients were --0.0180 and --0.0217 min/sup -1/. Hexobarbital catabolism was also accelerated above the control level when the mice were pretreated with hexobarbital and then tested 5 days after 700-r irradiation (elimination constant = --0.0170 min/sup -1/). Since the degradation of hexobarbital depends upon its demethylation and oxidation in liver microsomes, it was concluded that the organelles are injured by irradiation. (H.H.D.)
