ON THE MECHANISM OF CELL DESTRUCTION IN HEMATOPOIETIC ORGANS UNDER THE EFFECT OF IONIZING RADIATION

Mouse bone marrow and spleen (whole organs or fragments) were x irradiated with 700 to 5000 r and incubated in physiologic soiution for 4 hr at 37 un. Concent 85% C. In some experiments the organs from whole-body irradiated (700, 1000, 5000 r) mice were incubated and examined. In contrast to the evident injury of cells of irradiated intact animals, no destruction of cell elements in hematopoietic organs took place if cell elements were irradiated in vitro or if they were removed immediately after irradiation. However, bone marrow cells exposed in vitro to 700 r completely lost their ability to restore hematopoiesis in irradiated animals. No destructive changes were found in spleen cells if the spleen was surgically isolated or if its nerves and vessels were ligated 5 to 10 min after exposure of the animal to 1000 r. Removal of the ligature 1 to 3 hr later invariably led to the development of typical destructive processes in the organ. Cell suspensions irradiated with 1000 r in vitro and injected immediately after exposure directly into the spleen of unirradiated mice underwent destruction. The results indicate that radiosensitivity of celis of hematopoietic organs does not differ markedly in vivo and in vitro and that the destruction of hematopoietic elements may be interpreted in terms of direct radiation damage.
