RADIATION INJURIES IN THE PIG AND THEIR MODIFICATION BY AMINOETHYLISOTHIURONIUM CHLORIDE-HYDROCHLORIDE

Studies were performed to ascertain whether aminoethylisothiuronium chloride-hydrochloride (AET-HCl) provides protection against ionizing radiation in pigs; 300 pigs were used and x-ray doses of 500 to 1000 r. The LD/sub 50/30/ for the 5- to 7-week-old animals was found to be 720 r following unilateral irradiation in the dorsoventral direction. Increase in weight was disturbed by radiation in relation to the dose and attained a normal rate only after 3 to 4 weeks. Changes in the blood count showed the well-known marked decrease of leukocyte values, with slow recovery. Relative lymphocytosis and increase of young cells, both dependent on the dose, were observed. The onset of changes in the red blood count was slower. Serum protein values and albumin fraction were also reduced. Several pathologic changes were found. After tests of acute toxicity, AETHCI was given in doses of 100, 200, and 300 mg/kg orally before the onset of radiation. It improved the survival rate and gain in weight compared with controls submitted to the same radiation doses. The protection was not as marked as usually found with AET-HCI in tests on mice and rats: furthermore, no dependence on the dose of AET-HCI was seen. Blood count and blood serum tests revealed no protective action of AET-HCI. Lymphocytosis of the treated animals surpassed that of the controls. (TCO)
