ON THE MECHANISM OF THE THERAPEUTIC ACTION OF HIGHLY POLYMERIZED DNA IN RADIATION SICKNESS (in Russian)
To clarify the mechanism of action of highly polymerized DNA in acute radiation sickness, the fate of administered compounds in the normal and irradiated animals was studied directly, together with the effect of DNA on proliferation processes in bone marrow of irradiated animals. Fifty-five rats (there were 25 controls) were irradiated with 1000-r doses from Co/sup 60/, injected intraabdominally 24 hr later with 5 to 6 mg of highly polymerized DNA as liver, spleen, or thymus preparation in physiological saline, and 6 to 72 hr later were sacrificed. Homogenates of tissues were separated into DNA, RNA, and acid-soluble fractions. The DNA preparations, marked with P/sup 32/, were obtained from young rats partialiy hepatectomized and given NaHP/sup 32/O/sub 4/. The labeled DNA specific activity was highest in the liver in the first 6 hr after administration, 115% in the controls and 173% in the irradiated; it fell to 6% in both at 24 hr, and then rose at 72 hr to 70 and 30%, respectively. Spleen and bone marrow curves were somewhat similar in shape. The highest DNA activity was in the bone marrow in normal animals and in the liver in irradiated ones, and in most cases was lower in the irradiated than in the controls. In the bone marrow of the treated animals the mitotic activity was higher than in those not treated, and there were more nucleated cells. (TTT)
