EARLY CHANGES DETECTABLE BY ELECTRON MICROSCOPY IN THE BONE MARROW OF RATS AFTER WHOLE-BODY IRRADIATION (in German)

Intracellular structures in bone marrow cells were examined 30 to 60 min after exposure of rats to 500-r doses of Co/sup 60/ gamma radiation. Thirty min after irradiation, injury to these structures was clearly apparent. In eosinophil myelocytes, for example, mitochondrial swelling was evident and the cristae disorrented. Small cytoplasmic vacuoles were present and the Golgi zone was vesiculated. Otherwise, no structural changes were visible at this time. In plasma cells, vacuoles were present in the periphery of the Golgi zone, but the mitochondria and the well-developed endoplasmic reticulum appeared normal. However, marked structural changes occurred in reticulum cells at 30 min: mitochondria were markedly swollen and vacuolated with fragmented cristae. Although the membrane system was intact, the cytoplasm had a honeycomb appearance, and the nucleolr appeared granulated. At 1 hr after exposure the damage was more severe, especially in cells of the myeloid series. Granules in basophilic proinyelocytes were markedly degenerated and vacuolated. At this time defects in the plasma membrane were also present and vacuoles appeared under the nuclear membrane. From the observed morphologic injury to the mitochondria, it was concluded that their biochemical function in providing the cell energy metabolites would be impaired following irradiation. No decrease in the number of mitochondria per cell was noted in the 1-hr period. (BBB)
