EFFECT OF Pu$sup 239$ ON PHOSPHATASE ACTIVITY AND ON Ca, P, AND N CONTENTS OF BONE TISSUE IN RABBITS (in Russian)

Pathological processes occurring in bone tissue on entry of plutonium- 239 were studied by determining the phosphatase activity and the content of several substances in tissues of 84 rabbits. Plutonium nitrate solution was given intravenously, at the rate of 7 and 2 mu curie/kg, and animals were sacrificed after 3 to 180 days for the first dose and after 135 to 360 days for the second. Controls showed a decrease in the alkaline phosphatase activity in the epiphysis to 54% and in the diaphysis to 45%. Acid phosphatase activity also decreased significantly. The amounts of Ca, P, and total N in the bones did not charge statistically in either the controls or the animals given plutonium. However, in animals given 7 mu curie/kg plutonium, the alkaline phosphatase activity increased somewhat in 7 to 14 days after administration, but subsequentiy decreased and at 30 days was 70% of normal. Morphological observations indicated increased osteoblasts. After 90 and 135 days, the decrease in enzyme activity became clearer, and osteoblasts were significantly lower. After 180 days, alkaline phosphatase activity in the diaphysis was 61% of normal and in the epiphysis was 50%. After 30 days the acid phosphatase decreased to 37% in the diaphysis and to 28% in the epiphysis but did not change further. Morphological studies indicated high resorption with 2 mu curies/kg Pu. Alkaline phosphatase change were later, and only in the epiphysis, to 27% after 135 days. Osteoblasts increased, and the bone structure was atypical. (TTT)
