A total of 160 axolotls were given an x-ray dose of 8000 r to both hind extremities. Amputation of the legs was carried out 2.5, 3, and 6 months after the irradiation, and preparations containing RNA, DNA, or protein were injected into the right hind extremity or into the muscles of the back. The controls were not treated after amputation. Regeneration of the extremity took place in 13% of the cases with amputation 3 months after the irradiation, and in 23.3% of the cases with amputation 6 months after the irradiation. However, with repeated amputation, regeneration took place in 53 to 77% of the cases 6 months after the irradiation. Thus, the factor of shock was more important than the factor of time in restoring regenerative capacity. Commercial RNA and DNA were ineffective. High-molecularweight RNA prepared in the laboratory, and protein preparations were effective in restoring regenerative capacity of the extremities, but the RNA lost its activity on storage for a month and a half. Salamanders given a whole- body, x-ray dose of 1000 r and treated with RNA showed a 37% increase in life span as compared to the controls which were not treated with RNA. (TTT)
