Tests were conducted on the effects of protecting animals before irradiation with a mixture of 5 chemical agents, containing -SH groups or promoting anoxia, on certain serum components that may be related to natural immunity, such as normal antibody, lysozyme, and complement. Protection of guinea pigs or rabbits with this mixture against the lethal effects of x rays noticeably lessened the depressive effect of whole-body irradiation on normal serum levels of lysozyme, bacteriophage-neutralizing antibody, and bactericidal antibody. This was apparently caused by the so-called dosereducing effect of the agents. Neither protection nor irradiation seemed to have a significant effect on hemolytic complement titers of guinea pigs and rabbits. Reduction of a sublethal (500-r) x-ray dose to an estimated effective 250 r by the chemicals did not noticeably protect the immune antibody-forming mechanism of rabbits immunized during the radiosensitive stage of antibody formation. Protection against x irradiation afforded animals by the agents tested was attributed to several mechanisms. Diphenylamine, p-hydroxydiphenyl, and p-aminopropiophenone may protect tissue by lowering its oxygen content, whereas the sulfhydryl compounds cysteine and mercaptoethylamine may have the ability to trap free radicals.
