INFLUENCE OF IONIZING RADIATIONS ON THE EFFECTS OF HETEROLOGOUS IMMUNE CYTOTOXIN ON KB CELLS CULTURED IN VITRO (in Italian)

The combined effects of x rays and cytotoxic antibodies on KB cells were studied. Cytotoxic antiserum was prepared by subcutaneous injection of KB cells, emulsified in Freund adjuvant, in rabbits. When the cytotoxic antiserum with a titer of 1: 128 was incubated at a dilution of 1: 10 for 1 hr at 37 deg with 10/ sup 6/ KB cells in 5 ml of culture medium, only about 5% of the cells survived, as determined by their ability to form colonles. Survival was reduced to about 1% when the cells were exposed to a 100-r dose of x rays after incubation with cytotoxic serum and to approximates 0.5% when they were exposed to the same dose before treatment with immune cytotoxin. After irradiation only, approximates 40% of the cells survived. Only at doses above 1000 r was the metabolic activity of the cells appreciably depressed, as measured by acid production, but when smaller doses of radiation were combined with cytotoxin treatment, marked inhibition of metabolic activity was noted. This inhibition was greater when x- ray exposure preceded, rather than followed, cytotoxin treatment. Microscopic studies showed that after incubation with cytotoxin the cytoplasm of the cells became vacuolated and granulated, fragments of it broke off into the medium, and the nuclei became shrunken and pyknotic. All of these changes were intensified by concomitant exposure to x rays, particularly when irradiation preceded antiserum treatment. It is concluded that irradiation sensitizes the cells to damage by cytotoxic antibodies. (BBB)
