SPECTROPHOTOMETRIC METHOD FOR THE DETERMINATION OF LIPOXIDASE ACTIVITY: OBSERVATIONS ON UNSATURATED-FAT OXIDASE

BS>An improved spectrophotometric method for the determination of lipoxidase activity was developed and applied in studies of the purified enzyme and crude enzyme preparations from leguminous and other seeds, with linoleic acid as the substrate. The optimum pH was found to be crude lipoxidases from graham flour, soy beam meal, and hydrated mung beans, respectively. Hydrated mung beans showed two distinct modes of oxidation, the initial phase mediated by a nonspecific unsaturated-fat oxidase and the succeeding phase by lipoxidase. Brassica and lettuce seeds did not oxidize linoleic acid probably because of their antioxidant contents. Oxidation by wheat and barley seed extracts indicated that their activity was presumably due to the unsaturated-fat oxidase rather than lipoxidase. On the basis of the resistance of lipoxidase to enzyme inhibitors and ionizing radiation it is postulated that lipoxidase plays no role in the initiation of seed germination. The unsaturated-fat oxidase was found to be radiation sensitive, but its role in germination remains to be established. (auth)
