Granulopoiesis in the aplastic bone marrow of x irradiated rats after parabiosis with healthy litter-mate by aortic anastomoses

- For the purpose of revealing how the aplastic bone marrow of lethally irradiated animals restores hemopoietic activity after the anastomosis with healthy animals, the femural bone marrow of rats that received whole-body x irradiation 11,000 R), were parabiosed with healthy litter-mates by aontic anastomoses, and were sacrificed at certain intervals after parabiosis was observed. RNA and DNA synthesis was observed with /sup 3/H--Ur and /sup 3/H-- Tdr in vitro. By whole-body x irradiation with 1,000 R, all the immature cells in the bone marrow were eradicated by 3 days and the animals died 4 to 5 days after irradiation when the animals were left without parabiosis. But by parabiosis with healthy litter mates 3 days after irradiation, the hematopoietic activity in the bone marrow recovered completely on the third to founth day. A mass of mature granulocytes were settled in the aplastic bone marrow at least 6 hrs after parabiosis. Some of the granulocytes were enlarged and had basophilic cytoplasm. A few of myelocytea appeared after 12 hrs, and promyelocytes after 18 hrs. Mitoses were hardly observed by 12 hrs. The large sized granulocytes appearing after 6 hrs, showed RNA synthesis but not DNA synthesis. DNA synthesis began to oocur after 18 hrs in myelocytes and promyelocytes. The data suggest that granulocytes have the ability to dedifferentiate and probably acquire the character of younger procursor cells. (auth)
