Isolation and survival characterization of radiation and chemical-mutagen sensitive mutants of Pseudomonas aeruginosa

Using a variety of selection techniques, 52 radiation- and chemical mutagen-sensitive mutants have been isolated frcm Pseudomonas aeruginosa. These mutants have been phenotypically characterized and appear to constitute 12 groups separable on the basis of their survival responses and phage reactivation ability. The majority of the mutants are sensitive to uv radiation and/or methyl methanesulfonate (MMS). There appears to be cross sensitivity between MMS and nitrosoguanidine (NTO), but not between either of these and gamma radiation. The presence of host-cell reactivation deficient mutants for phages treated with uv light or MMS suggests that the lesions induced by these can be repaired to some extent in the pareni strain, whereas the lack of host-cell reactivation (HCR) for gamma -irradiated phages could mean that there is no repair of this type of damage. Four classes of mutant that do not yet have their parallels among the radiation-sensitive mutants of other bacterial species are reported. (auth)
