Specificity of solid phase radioimmunoassay of Australia antigen and anti- Australia antigen (in Japanese)

The specificity of solid-phase radioimmunoassay method for Australian antigen (Au-ag) and anti-Australian antigen (Au-ab) was confirmed. Au-ab was coated quantitatively on the surface of /sup 125/I labeled Au-ag. The /sup 125/I- labeled Au-ag showed positive reaction by using a tube coated with Au-ab, but negative by using tube coated with normal sera or any other kinds of antibody. The combination of /sup 125/I(-labeled Au-ab complex that was formed on the surface of a coated tube was blocked with unlabeled Au-ab. When normal human serum, several kinds of animal sera, several viruses, enzymes, and hormones were estimated in solid phase radioimmunoassay (SP-RrA), no increase of radioactivity was observed. When Au-ag or Au-ab was quantitatively detected by SP-RIA, the cutoff value to be accepted as a positive value should be beyond seven standard deviations from the negative control serum cpm. The arbitrary unit of Au-ag and Au-ab could be obtained from the estimated value in SP-RIA. (auth)
