In vitro and in vivo labeling of rabbit blood leukocytes with $sup 67$Ga- citrate

The labeling characteristics of rabbit blood leukocytes with /sup 67/ Gacitrate, both in vitro and in vivo, have been defined. Both granulocytes and lymphocytes were consistently labeled by the in vitro incubation of anticoagulated whole blood with /sup 67/Ga-citrate, Minimal uptake by red blood cells occurred. Both the absolute and relative uptake of the radionuclide by granulocytes as compared with lymphocytes was greater in media-1640 than in anticoagulated whole blood. Maximal uptake of label by leukocytes occurred within 1 hr, was independent of temperature of incubation and type of anticoagulant, and occurred at a concentration of approximately 200 mu Ci /dup 67/Ga/10 ml of heparinized blood. Comparative studies with /sup 51/Cr demonstrated that /sup 67/Ga produced a label on both granulocytes and lymphocytes equal to or greater than the label obtained with /sup 67/Cr and that there was minimal loss of label by /sup 67/Ga-labeled cells with in vitro incubation. Blood leukocytes labeled in vitro with /sup 67/Ga offer an excellent gamma-emitting radioactive tracer that can be used to develop a technique for the scintigraphic detection and localization of abscesses.
