Radioimmunoassay of digoxin (in Japanese)

An artificial antigen combined with bovine serum albumin (BSA) and digoxin was used, because digoxin has a low molecular weight and no antibody producing capacity. An artificial antigen was administered to a rabbit, and after 30 to 70 days an antiserum of high antibody value was obtained. This serum was strongly specific to digoxin, especially to the aglycon fraction. /sup 3/H- digoxin was used as labeled-antigen, and the dextran-coated charcoal method was used. The relationship between a dose of digoxin (maintenance dose) and the concentration of digoxin in the blood was studied in 113 cases. and the level in the blood. In cases with renal insufficiency with blood became far higher. In the methods for digitalis measurement in the blood such as bioassay, Rb, the double isotope dilution derivative assay, rndioimmunoassay with /sup 125/I- digitoxin, and enzymatic assay, the principle, the sensitivity, and other problems were compared. RIA in the measurement of digoxin was a superior method for clinical investigation, in which low concentrations of digoxin in the blood could be more simply measured in a short time. (JA)
