The effects of whole-body exposure to x radiation doses of 350 r in mice, 350 r in young rats, and 180 to 200 r in young guinea pigs on the blood picture were followed for 90 days following irradiation. Phosphorus-32 was used as a tracer in studies of spleen metabolism. Both medullary hematopoiesis in various bones and extramedullary hematopoiesis in the spleen were correlated with histological evidence of radiation injury. It was concluded that the increase in spleen weight beyond normal that was observed to reach a peak 20 days post- irradiation was due to the vigorous acceleration of extramedullary hematopoiesis. This response was considered to be compensatory in nature in view of the inadequacy of bone marrow hematopoiesis during this period, and it was concluded it was responsible for the sudden increase in neutrophils observed in peripheral blood at this time.
