ESTABLISHMENT AND STABILITY OF CHIMERISM BETWEEN TWO CLOSELY RELATED LINES OF MICE

Skin grafts and isografts were exchanged between two strains of mice, CBA and CBA/T6T6 (CBA containing T6 marker chromosome). All interstrain grafts between CBA and CBA/T6T6 were rejected but with prolonged rejection times, indicating that the two strains do not differ at the H-2 locus. CBA/T6T6 spleen cells were injected intravenously into new-born CBA mice; all injected mice survived and were tolerant to CBA/T6T6 skin grafts 6 weeks after injection, but no CBA/T6T6 cells were found in the spleen, bone marrow, or thymuses. CBA mice were irradiated and then injected with CBA/T6T6 bone marrow, and at intervals some of the mice were subjected to cytological analysis. Donor cells were found to dominate at all doses, and no host cells were seen after 700 r. The stability of the chimeras is discussed. (D.L.C.)
