Simultaneous inhibition of thymidine degradation and stimulation of incorporation into DNA by 5-fluorodeoxy-uridine

Cultured plant cells degrade exogenously supplied thymidine. 5-Fluorode- oxyuridine (FdUrd) can be used to inhibit this degradation while at the same time enhancing thymidine (dThd) incorporation into DNA. If $sup 14$C- or $sup 3$H- labeled dThd is used, the improvement in DNA labeling which results permits detection of DNA synthesis in freshly isolated protoplasts, as well as other sensitive measures of DNA metabolism in plant cells.
