The human Gly R {alpha}{sub 1} subunit cDNA was labeled by nick-translatoin using biotin-14-dATP and hybridized in situ at a final concentration of 20 ng/{mu}l to metaphase chromosome spreads from two normal males. The FISH method was modifed from that previously described in that chromosomes were stained with both propidium iodide (as a counterstain) and DAPI (for chromosome identification) before analysis. Images of metaphase preparations were caputed by a CCD camera and were computer enhanced.
