LDMS for fast DNA analysis and disease diagnosis

Since the discovery of matrix-assisted laser desorption (MALD) for protein detection by Hillenkamp and his co-workers, matrix-assisted laser desorption mass spectrometry has been successfully used to detect various kinds of proteins. Proteins with molecular weights higher than 300,000 amu have been detected. However, when MALD was applied to DNA, the success was limited to small DNA segments. Recently, by using a custom designed time-of-flight mass spectrometer with high ion energy combined with specific mixtures of organic compounds as matrices, the authors succeeded in detecting oligonucleotides up to 500 base pairs in size. The authors succeeded in detecting the codon 508 mutation in cystic fibrosis carriers and patients.
