Studies of individual large molecules by ESI-FTICR mass spectrometry

The authors have developed methods for molecular weight (MW) determination of megadalton molecules based on the measurements of individual ions by FTICR-MS. Stepwise charge state shifts of individual ions in reaction with selected reagents produce multiple peaks from the same ion, from which charge states and MW can be calculated. Alternatively, the number of charges can be measured simultaneously with the mass-to-charge ratio to obtain the molecular weight of an individual ion, although with precision currently limited to 5 to 10%. For the case of very large molecules (>10 MDa), only the latter approach has been effective thus far.
