Isolation of candidate tumor suppressor genes for small cell lung cancer (SCLC) from human chromosome 3p21.3

Lung cancer is the leading cause of cancer related deaths in the United States. SCLC tumors frequently display deletions on the short arm of chromosome 3 suggesting the existence of a tumor suppressor gene in this region. The region 3p21.3 is one of the common regions of deletions in lung cancer as shown by loss of heterozygosity studies and deletion mapping data and more recently by occurrence of several homozygous deletions in the same region. In our laboratory we have discovered 3 overlapping homozygous deletions in SCLC at 3p21.3. The region has been covered in YAC, p1 and cosmid contigs. Here we describe the isolation, characterization and mutational analysis of several candidate genes for SCLC in the homozygously deleted region at 3p21.3.
