Title: Single crystal x-ray study of leucyl-tRNA-aminoacyl-ligase using an image intensifier-aided detection system

This thesis reports on the successful application of a high speed X-ray recording system to the study of leucyltRNA- aminoacyl-ligase crystals from baker's yeast. This protein has proved difficult to study using conventional . techniques because of its limited supply and weak diffraction pattern. The recording system used a rotation camera geometry. The X-rays were converted into light by a thin phosphor; this light was amplified by a high gain, magnetically focused image intensifier, and the diffraction pattern was recorded on photographic film. Complete 3 Å data sets were collected from one native protein crystal and from one gold derivative crystal. The recording of each complete set took approximately sixteen hours. The mean discrepancy between symmetry-related reflections was rather high, 20% to 25%. However, most'unique reflections were observed many times, and the standard deviation of their mean intensities averaged 15%. These intensities - were used to compute a difference Patterson map for the gold derivative and possible heavy-atom sites were located. The apparatus described in this thesis is therefore capable of collecting large amounts of useful X-ray data from very few protein crystals.
