Genotoxicities of organic extracts of airborn particulates in SOS Chromotest and Ames test

The rapid development of transportation industries, and other fuel consumption resulted in a quite serious air pollution in Guangzhou, China, as revealed by a WHO-supported air pollution monitoring program. Meanwhile, the incidence of lung cancer increased in the past decade, air pollution being considered as a possible factor in the causes of the cancer. We carried out studies following the aforementioned program, using two bacterial testing systems to examine the genotoxicities of the organic extracts of the airborn particulates were collected on fiberglass filters in a high-volume sampler in 4 districts in Guangzhou, and then extracted in Soxhlet extractors with methylene chloride for 4-8hours, condensed, weighed and dissolved in DMSO. SOS Chromotest (tester strain PQ37) and Ames Test (tester strains TA98 and TA100) were performed without S9 mix. The extracts showed positive results in the two tests, the lowest concentrations induced positive reaction being 1 mg/ml and 2.5 mg/ml respectively. The genotoxicities of the organic extracts of the particulates of different seasons were compared, but no significant difference was found. It was found that the storage of the extracts at 0{degrees}C had a slight influence on the genotoxicities.
