The cytogenetic analysis of mice chronically fed PhIP or MeIQx

The cytogenetic effects in mice chronically fed heterocyclic amines were evaluated by chromosome painting and SCEs. Two such compounds, PhIP and MeIQx, have been isolated from cooked foods and found to be carcinogenic in laboratory rodents. Female C57Bl/6 mice were chronically fed diets containing 0, 100, 250 or 400 ppm of PhIP or MeIQx beginning at 8 weeks of age. Peripheral blood and bone marrow were taken from 5 mice per chemical and dose 1, 2, 4, and 6 months from the start of exposure. Chromosome-specific composite DNA probes for chromosomes 2 and 8 were hybridized to metaphase spreads isolated from each tissue. The 1 and 4 month time points show no significant difference in chromosome aberration frequencies between the control mice and the mice exposed at any dose of either chemical. At approximately the 4 month time point, mice exposed to 400 ppm PhIP began exhibiting signs of toxicity including weight loss. SCEs were visualized following 6 months (PhIP) and 7 months (MeIQx) exposure. Analysis of SCEs in both tissues of MeIQx mice are in progress. Results from the peripheral blood of mice exposed to PhIP show an approximate doubling compared the control mice while the bone marrow shows no significant difference between exposed or control mice.
