Agar plate cultures are discussed with regard to preparation of agar plates and isolation of clones, special considerations for agar plate culture, and colony formation of various cell lines on agar plates. Methods of Lederberg-style replica plating of mammalian cells are described. Applications of algar plate culture and replica plating to mammalian cell genetics are discussed with regard to isolation of drug-resistant clones using agar plates, isolation of auxotrophic mutants, and isolation of uv-sensitive clones.
