Effects of hypoxia on human lymphocyte activation

Peripheral blood mononuclear cells were obtained from 7 subjects at sea level (SL), 7500 ft (Alt/sub 1/) and 25,000 ft (Alt/sub 2/) during a 7 week decompression chamber study. Circulating leukocyte subpopulations were identified by flow cytometry using fluoresceinated monoclonal antibodies. In vitro PWM and PHA stimulated /sup 35/S Methionine incorporation was determined at 24 hours, /sup 3/H Thymidine uptake at 72 hours, and NK cytotoxicity at 24 hours by /sup 51/Cr released from K562 cells. An increase in percentages of monocytes (9 +/- 1 SL vs. 19 +/- 3 Alt/sub 2/) was observed without changes in T helper, T suppressor or B cells. NK cytotoxicity was unimpaired in 6 of 7 subjects at Alt/sub 2/ despite reduced PHA but not PWM stimulated protein synthesis at 24 hours and reduced blastogenesis at 72 hours. This study indicates that hypoxia may impair host resistance in humans by inducing immunoregulatory abnormalities.
