Model fluorogenic reagents have been synthesized and evaluated toward the goal of ultrahigh-sensitivity chromatographic analysis of primary amines with laser-based detection. Chemical reactivity and spectral criteria were utilized to evaluate 12 potential reagents forming highly fluorescent isoindoles. 3-Benzoyl-2-quinolinecarboxaldehyde has been chosen at this time as the most promising reagent for the precolumn derivatization of model amino acids, followed by a chromatographic analysis of the formed isoindoles, and their detection with a helium-cadmium laser-based detector at the femtogram level.
