Urinary kininogen excretion in spontaneously hypertensive rats (SHR) determined by a new radioimmunoassay

Plasma low mol. wt. kininogen (LMWK) was isolated to apparent homogeneity and antibodies raised in rabbits. An RIA was developed with a standard curve of 2.5-100 ng. LMWK was /sup 125/T-labelled at 2-4 ..mu..ci/..mu..g. Displacement of tracer from antibody by 1-10 ..mu..l of urine from four rats was parallel. In 28 tests interassay C.V. of a rat urine pool was 12.4%. Intra-assay C.V. in 11 tets was 9.4%. In 18 expts. recovery of pure LMWK was 110.0 +/- 3.40%. In twenty urines, kininogen was analyzed by trypsin digestion and by the RIA with r/sup 2/=1.00. Cross-react. of bovine LMWK was 2.5% and that of rat HMWK (Dr. Alhenc-Gelas) was 5.8% (both non-parallel). Bradykinin, 200 ng/tube, did not show any apparent displacement. LMWK excretion and kallikrein (KK) were determined in 10 Wistar Kyoto (WKY) and 9 SHR. The following conclusions were reached: (1) KK is reduced in SHR as previously reported; (2) they now report that LMWK excretion is significantly increased in SHR. The increase of LMWK in SHR may be due to reduced consumption because of lower KK. Also, glomerular damage due to hypertension may have increased filtration of LMWK.
