Assay for the cholecystokinin receptor on pancreatic acini

For binding studies with cholecytokinin receptors, radiolabelled hormone of moderately high specific activity must be prepared. A satisfactory radiolabelled peptide can be obtained by labelling pure porcine cholecystokinin with radioiodinated Bolton-Hunter reagent. When dispersed acini are incubated with (/sup 125/I(cholecystokinin at 37/sup 0/, binding of the labelled peptide reaches a steady-state by 45 min. Bound (/sup 125/I)cholecystokinin dissociates slowly. The rate of dissociation is temperature dependent and is increased by 50% by adding unlabelled cholecytokinin.
