Quantification of platelet thrombosis on bovine pericardial valve prostheses and reduction of thrombosis with diphosphonate treatment in calves

Effect of calcification-inhibitor, sodium diphosphonate on thrombosis on bovine pericardial valve prostheses was evaluated in 4-week-old Holstein calves with In-111 labeled platelets. Twenty-two calves, 11 control, 11 diphosphonate treated (5 mg/kg/day, subcutaneously) were implanted with 25 mm bovine pericardial valve prostheses in mitral annulus and killed 30 and 90 days post-implantation. Twenty-four hours before killing 350-450 ..mu..Ci of autologous In -111 platelets were administered I.V. Components of valve were separated, weighed and area determined. Mean (+-SD) value of platelet deposition (platelet x 10/sup 3//mm/sup 2/) on four sections of leaflet (free edge, central zone, flexion zone, and attachment zone) was calculated from platelet count, radioactivity in blood, leaflet sections and area. In addition to reduction of calcification, diphosphonate also reduces platelet thrombosis in tissue valve prostheses. Platelet deposition at free edge increased at 90 days due to rough calcified surface in control calves.
