Photoaffinity labeling of prolactin receptors

In order to elucidate the molecular characteristics of prolactin (PRL) receptors, a direct electrophoretic analysis of the relative molecular mass (M/sub r/) of purified PRL receptors and an electrophoretic analysis of photoaffinity-labelled receptors was utilized. Results of the study indicate that the major binding component in the membrane of rabbit mammary tissue has a M/sub r/ of 31,000-32,000 when affinity labelling combined with SDS-polyacrylamide gel electrophoresis were employed. Using electrophoresis under reducing conditions, a single M/sub r/ of 58,500 binding component-(/sup 125/I)PRL complex was obtained from rabbit mammary gland microsomes. Similar M/sub r/ binding components were also observed for other tissues from rabbits and pig mammary glands and slightly higher in M/sub r/ components from rat liver.
