The distribution and metabolism of Tc-/sup 99m/ labeled low density lipoprotein in human subjects

The distribution of Tc-99m labeled low density lipoprotein (LDL) was determined in 8 human subjects with documented atherosclerosis involving the carotid or coronary arteries. Autologous LDL was isolated by ultracentrifugation, labeled with 370 to 740 MBq of Tc-99m using a dithionite reduction technique, purified on a column of Sephadex G-50 and administered intravenously. Serial images were recorded over a 28 hour interval using a large field scanning scintillation camera and a dedicated computer. In parallel with the imaging, serial blood samples were obtained to define blood clearance. The data were displayed to identify sites of focal tracer concentration in the arterial tree and to define changes in heart and liver activity. The radiopharmaceutical had excellent blood pool retention, with remarkable contrast between blood pool and soft tissues throughout the imaging interval. The half-time of clearance of LDL from blood was 29.66 +- 8.98h which was in good agreement with the value of 27.67 +- 6.72h determined by imaging.
