The sensitivities of six human cell lines to 50-MeV neutrons were studied using the colony-forming assay in vitro. Two models were used to fit the survival curves: the linear-quadratic (L-Q) model and the single-hit multitarget model with initial slope. The quality of the resulting fits was compared with two analytical procedures. One measured the accuracy of the fits, and the other led to an estimation of the validity of the parameters obtained. The L-Q model responded satisfactorily to these two procedures and could be used to demonstrate that: (1) neutron action can be explained adequately by the same model of cell killing that applies to ..gamma.. rays; (2) neutron survival curves were different for each cell line; (3) the neutron survival parameters change differently for each line in comparison to ..gamma..-ray survival parameters; and (4) the biological parameters determining the differences among cell lines are of utmost importance in determining radiosensitivities at high and low linear energy transfers. Relative biological effectiveness varied from 3 to 1.2.
