Comparison of radiolabeled monoclonal antibodies in an in vitro tissue binding assay
The authors previously described a simple in vitro assay for evaluating radiolabeled monoclonal antibodies (Abs) as an aid in selecting for clinical trials those with superior tissue binding properties. Tumor or normal tissues as 1 mm cubes are incubated in microtitre-plate wells containing media and indium-111 labeled Abs and, following rinsing, are counted to measure binding. The assay has now been used to evaluate 4 anti-colorectal tumor Abs against 6 human colorectal tumor tissues and normal liver and colon with an additional Ab (anti-PAP) and tissue (CX-1, an established colorectal tumor) as controls. In addition, 4 anti-breast tumor Abs were evaluated against 7 human tumor tissues and normal tissues from 8 organs. Differences in binding of one Ab to different tissues and of different Abs to one tissue were observed in both studies which were usually for larger than the range of repeat measurements. Lower binding was usually observed for the control Ab and for normal tissues. However, increased binding of anti-tumor Abs occasionally occurred in normal tissues especially in spleen and pancreas tissue with anti-breast tumor Abs and in normal colon tissue with one anti-colorectal tumor Ab. A comparison with immunoperoxidase staining in the same tissues with the anti-breast tumor Abs showed a pattern of specificity not dissimilar from that shown by the assay.
