Development of radioimmunoassay for thromboxane B/sub 2/

A sensitive and specific radioimmunoassay for thromboxane B/sub 2/ has been developed. Thromboxane B/sub 2/ antiplasma was produced in rabbits by repeated immunization with thromboxane B/sub 2/ coupled to bovine serum albumin. The (/sup 125/I)-labeled antigen with high specific activity was prepared by radioiodination of thromboxane B/sub 2/-tyrosine methyl ester conjugate followed by purification with thin-layer chromatography. Thromboxane B/sub 2/ antiplasma cross-reacted less than 0.05% with PGD/sub 2/ or PGF/sub 2..cap alpha../ and less than 0.01% with other prostaglandins, arachidonic acid, or endoperoxide analogs. The sensitivity of the assay was 5 pg per assay tube.
