Hen erythrocytes were ..gamma..-irradiated (up to 15 kR) and incubated at 37/sup 0/C for 20 hr in a slightly hypotonic saline solution (0.10 M NaCl buffered with 5 mM phosphate, pH 7.4) to which 2,4-dinitrophenol, glucose, NaF, or pyruvate were added in different combinations. Hemolysis was measured at the end of the incubation period. Dinitrophenol (2 x 10/sup -4/ M) reduced the radiation-induced hemolysis in both ATP-containing and ATP-depleted erythrocytes. Glucose (10 mM) and pyruvate (10 mM) antagonized the protective effect of dinitrophenol against radiation-hemolysis. NaF suppressed the antagonistic action of glucose on the protective effect of dinitrophenol.
