Evaluaion of a nuclease-resistant derivative of poly(I) poly(C) (poly(ICLC)) as a radioprotective agent. [Whole-body. gamma. -irradiated mice]

Intramuscular injection of a stabilized polyriboinosinic acid-polyribocytidylie acid complex, composed of the two acids poly-l-lysine and carboxymethylcellulose (poly(ICLC)), was radioprotective in mice when given in single or multiple doses before total-body irradiation. At low to moderately high dosages, LD/sub 50/30/ values were increased significantly (P < 0.05), with a maximum dose-reduction factor of 1.25. Survival rates and weighted mean survival times were also significantly increased. Increased circulating levels of serum interferon proportional to poly (ICLC) concentration were not required at the time of irradiation for radioprotection. Poly (ICLC) was found to have a slight stimulatory effect on the numbers of peripheral blood elements in irradiated mice.
