Use of preparative chromatography to isolate pyridine bases
The ..beta..-picoline fraction derived from coal tar contains 18 to 20% ..cap alpha..-picoline, 20 to 32% 2,6-lutidine, 30 to 32% ..beta..-picoline, and 28 to 30% ..gamma..-picoline. It provides a feedstock for the isolation of pure compounds used to produce vitamins and medicinal preparations. At present, pure ..beta..- and ..gamma..-picoline are in most demand. The difficulty of separating them by rectification arises from the small b.p. difference (they boil at 144.1 and 145/sup 0/C, respectively). Methods of separation include fractional freezing out, azeotropic rectification, complex formation with metal salts, and other complicated methods which have not been developed beyond the laboratory state. Preparative chromatography is an interesting alternative method of isolating ..beta..-picoline from the ..beta..-picoline fraction. The chromatographic procedure is sufficiently flexible to overcome numberous problems in the analytical and preparative separation of mixtures for the purposes of quantitative determination or the isolation of specified constituents. A method of preparative chromatography is described for the isolation of individual and mixed pyridine bases.
