The response to hyperthermia, both alone and in combination with x irradiation, was examined in Chinese hamster V-79 cells in vitro. The effect of hyperthermia in the range from 40 to 45/sup 0/C on cell survival differed markedly at temperatures below 42/sup 0/C and above 43/sup 0/C. Preheating the cells for 2 hr at 42/sup 0/C slightly decreased their sensitivity to the subsequent exposure to 44/sup 0/C, whereas a 15-min exposure to 44/sup 0/C markedly increased the response of the cells to 42/sup 0/C. When cells were exposed to x irradiation in combination with heating at 44/sup 0/C for 15 min and 42/sup 0/C for 60 min, the radioenhancing effect was larger with the 44 ..-->.. 42/sup 0/C sequence than with the 42 ..-->.. 44/sup 0/C sequence, regardless of the position of x irradiation in the sequence of the combined treatments. From these results, it seems that the hyperthermic damage by exposure to 44/sup 0/C may be different from that by exposure to 42/sup 0/C. For the application of hyperthermia to radiotherapy, it is suggested that a short hyperthermic treatment at 44/sup 0/C followed by a long treatment at 42/sup 0/C in combination with x irradiation may be an efficient protocol.
